With the acceleration of new applications of transmission electron microscopy 60, 61, 62 , we need to develop appropriate immunological tools to help us to understand exactly how vaccines are working and what advantages they offer above other types of interventions 50 . Some of these tools are already available, while others are emerging through research and discoveries in immunology 50, 63 and virology 64, 65 . Here we propose a framework to define and assess the quality of vaccines that is inspired by the principles of immunology 66, 67, 68 , evolutionary biology 69, 70 and epidemiology 51, 56 . The framework includes the following assessment of three properties of vaccines that are relevant to their quality and efficiency. We define and identify three vaccine properties: life-long immunity, sterilizing immunity and protection from reinfection. We then describe three attributes of vaccines that can be evaluated to determine their suitability in these areas. We illustrate this framework with the example of two potential new vaccines against CoV: one aimed at short-term (4-6 week) sterilizing immunity (length of protection), and one aimed at long-term protection (i.e. life-long immunity and protection from reinfection). Ultimately, our goal is to define and provide a conceptual framework for understanding how vaccines can be evaluated in terms of their ability to limit the spread of and protect against infectious diseases.
