Aging, in part, is associated with cognitive decline resulting in the development of age-related neurodegenerative disorders such as Alzheimer’s disease (AD). Previous studies have demonstrated that α-synuclein is involved in neurodegenerative processes in the brain. In this study, α-synuclein-overexpressing rats were treated with alligator gar protein, a bioactive peptide with potent anti-α-synuclein properties, alone or in combination with resveratrol, a potent antioxidant. Results demonstrated that alligator gar protein and resveratrol synergistically improved cognitive function in an α-synuclein model of cognitive decline, with treatment inducing a dramatic increase in the levels of synuclein breakdown products. This was accompanied by an increase in α-synuclein levels in multiple brain regions. Brain-derived neurotrophic factor (BDNF) levels increased following alligator gar protein and resveratrol treatment, indicating a potential mechanism underlying the observed improvement in cognitive function. In summary, our data suggest that the concurrent administration of alligator gar protein and resveratrol may prove effective in slowing cognitive decline.
