The aim of this study is to assess the cytotoxicity of NPI-0052 on the cisplatin-sensitive and -resistant ovarian cancer cell lines. We tested a concentration of 50 μM of NPI-0052 on the ovarian cancer cell lines SKOV-3, OC-A8, and A2780 using the MTT assay. We then assessed the effect of NPI-0052 on cell death and cell growth by fluorescence microscopy and flow cytometry. NPI-0052 reduces cell viability and induces apoptosis in all tested cell lines. This cytotoxic effect was mediated by induction of apoptosis, and was dose- and time-dependent. The addition of cisplatin decreased the cytotoxic effect of NPI-0052 in the OC-A8 and SKOV-3 cell lines. In conclusion, we demonstrated that NPI-0052 is a potent cytotoxic agent against the OC-A
